Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001005242.3(PKP2):c.16G>A (p.Ala6Thr), citing Ambry Variant Classification Scheme 2023: The p.A6T variant (also known as c.16G>A), located in coding exon 1 of the PKP2 gene, results from a G to A substitution at nucleotide position 16. The alanine at codon 6 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
