Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001430.5(EPAS1):c.1658C>A (p.Ala553Glu), citing Ambry Variant Classification Scheme 2023: The p.A553E variant (also known as c.1658C>A), located in coding exon 12 of the EPAS1 gene, results from a C to A substitution at nucleotide position 1658. The alanine at codon 553 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
