Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_025137.4(SPG11):c.1657G>A (p.Glu553Lys), citing Ambry Variant Classification Scheme 2023: The p.E553K variant (also known as c.1657G>A), located in coding exon 8 of the SPG11 gene, results from a G to A substitution at nucleotide position 1657. The glutamic acid at codon 553 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
