Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.1051C>T (p.His351Tyr), citing Ambry Variant Classification Scheme 2023: The p.H351Y variant (also known as c.1051C>T), located in coding exon 10 of the RAD54L gene, results from a C to T substitution at nucleotide position 1051. The histidine at codon 351 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
