NM_000302.4(PLOD1):c.1631_1632delinsGC (p.Ala544Gly) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1631_1632delCAinsGC variant (also known as p.A544G), located in coding exon 15 of the PLOD1 gene, results from an in-frame deletion of CA and insertion of GC at nucleotide positions 1631 to 1632. This results in the substitution of the alanine residue for a glycine residue at codon 544, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
