NM_007194.4(CHEK2):c.1624G>A (p.Val542Met) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V542M variant (also known as c.1624G>A), located in coding exon 14 of the CHEK2 gene, results from a G to A substitution at nucleotide position 1624. The valine at codon 542 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
