NM_005477.3(HCN4):c.1623C>A (p.His541Gln) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H541Q variant (also known as c.1623C>A), located in coding exon 5 of the HCN4 gene, results from a C to A substitution at nucleotide position 1623. The histidine at codon 541 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
