NM_001267550.2(TTN):c.43374T>A (p.Asp14458Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D5393E variant (also known as c.16179T>A), located in coding exon 62 of the TTN gene, results from a T to A substitution at nucleotide position 16179. The aspartic acid at codon 5393 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and glutamic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
