NM_007194.4(CHEK2):c.1606C>T (p.Pro536Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P536S variant (also known as c.1606C>T), located in coding exon 14 of the CHEK2 gene, results from a C to T substitution at nucleotide position 1606. The proline at codon 536 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
