Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.1601A>C (p.His534Pro), citing Ambry Variant Classification Scheme 2023: The p.H534P variant (also known as c.1601A>C), located in coding exon 3 of the TERT gene, results from an A to C substitution at nucleotide position 1601. The histidine at codon 534 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
