NM_007194.4(CHEK2):c.1598del (p.Thr533fs) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1598delC variant, located in coding exon 14 of the CHEK2 gene, results from a deletion of one nucleotide at nucleotide position 1598, causing a translational frameshift with a predicted alternate stop codon (p.T533Kfs*33). This alteration occurs at the 3' terminus of the CHEK2 gene, is not expected to trigger nonsense-mediated mRNAdecay and results in the elongation of the protein by 21 amino acids by the removal of the last 11 native amino acids and the addition of 32 alternative amino acids. The exact functional effect of the altered amino acids is unknown. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
