Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.1591A>C (p.Lys531Gln), citing Ambry Variant Classification Scheme 2023: The p.K531Q variant (also known as c.1591A>C), located in coding exon 15 of the PLOD1 gene, results from an A to C substitution at nucleotide position 1591. The lysine at codon 531 is replaced by glutamine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
