Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_170707.4(LMNA):c.1577_1582dup (p.Arg527_Thr528insMetArg), citing Ambry Variant Classification Scheme 2023: The c.1577_1582dupTGCGTA variant (also known as p.R527_T528insMR), located in coding exon 9 of the LMNA gene, results from an in-frame duplication of TGCGTA at nucleotide positions 1577 to 1582. This results in the insertion of 2 extra residues (methionine and arginine) between codons 527 and 528. This amino acid region is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
