This variant lies in the BARD1 gene (transcript NM_000465.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1568, duplicating one base. Submitter rationale: This variant causes a duplication of one nucleotide at the +2 position in intron 6 of the BARD1 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with BARD1-related disorders in the literature. This variant has been identified in 1/251120 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Although there is a suspicion that this variant may be associated with disease, additional studies are necessary to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
