Uncertain significance — the classification assigned by Ambry Genetics to NM_052947.4(ALPK2):c.1561G>T (p.Gly521Trp), citing Ambry Variant Classification Scheme 2023: The p.G521W variant (also known as c.1561G>T), located in coding exon 3 of the ALPK2 gene, results from a G to T substitution at nucleotide position 1561. The glycine at codon 521 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
