NM_000492.4(CFTR):c.1561A>C (p.Ile521Leu) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the CFTR gene (transcript NM_000492.4) at coding-DNA position 1561, where A is replaced by C; at the protein level this means replaces isoleucine at residue 521 with leucine — a missense variant. Submitter rationale: The p.I521L variant (also known as c.1561A>C), located in coding exon 11 of the CFTR gene, results from an A to C substitution at nucleotide position 1561. The isoleucine at codon 521 is replaced by leucine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
