Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004329.3(BMPR1A):c.1558A>G (p.Thr520Ala), citing Ambry Variant Classification Scheme 2023: The p.T520A variant (also known as c.1558A>G), located in coding exon 11 of the BMPR1A gene, results from an A to G substitution at nucleotide position 1558. The threonine at codon 520 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
