Submitter rationale: The p.K520E variant (also known as c.1558A>G), located in coding exon 16 of the DNAI1 gene, results from an A to G substitution at nucleotide position 1558. The lysine at codon 520 is replaced by glutamic acid, an amino acid with similar properties. This variant has been detected in conjunction with a pathogenic mutation in DNAI1 by our laboratory in trans. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is conserved in all available species, except gorilla, medaka and lamprey. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
