NM_002907.4(RECQL):c.1550A>G (p.Asp517Gly) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D517G variant (also known as c.1550A>G), located in coding exon 12 of the RECQL gene, results from an A to G substitution at nucleotide position 1550. The aspartic acid at codon 517 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
