Uncertain significance for Dilated cardiomyopathy 1J — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004100.5(EYA4):c.1549G>A (p.Glu517Lys), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with EYA4-related conditions. This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 517 of the EYA4 protein (p.Glu517Lys). This variant is not present in population databases (gnomAD no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
