Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_030777.4(SLC2A10):c.1546C>T (p.Arg516Trp), citing Ambry Variant Classification Scheme 2023: The p.R516W variant (also known as c.1546C>T), located in coding exon 4 of the SLC2A10 gene, results from a C to T substitution at nucleotide position 1546. The arginine at codon 516 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species; however, tryptophan is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
