Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.1544T>C (p.Leu515Pro), citing Ambry Variant Classification Scheme 2023: The p.L515P variant (also known as c.1544T>C), located in coding exon 12 of the RECQL gene, results from a T to C substitution at nucleotide position 1544. The leucine at codon 515 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
