Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.1542A>C (p.Glu514Asp), citing Ambry Variant Classification Scheme 2023: The p.E514D variant (also known as c.1542A>C), located in coding exon 11 of the CFTR gene, results from an A to C substitution at nucleotide position 1542. The glutamic acid at codon 514 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
