Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.1540A>G (p.Lys514Glu), citing Ambry Variant Classification Scheme 2023: The p.K514E variant (also known as c.1540A>G), located in coding exon 12 of the RECQL gene, results from an A to G substitution at nucleotide position 1540. The lysine at codon 514 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
