NM_000251.3(MSH2):c.1535A>C (p.Lys512Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K512T variant (also known as c.1535A>C), located in coding exon 10 of the MSH2 gene, results from an A to C substitution at nucleotide position 1535. The lysine at codon 512 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species.
