Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001256715.2(DNAAF3):c.1293G>T (p.Arg431Ser), citing Ambry Variant Classification Scheme 2023: The p.R498S variant (also known as c.1494G>T), located in coding exon 12 of the DNAAF3 gene, results from a G to T substitution at nucleotide position 1494. The arginine at codon 498 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
