Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.1483A>G (p.Ser495Gly), citing Ambry Variant Classification Scheme 2023: The p.S495G variant (also known as c.1483A>G), located in coding exon 10 of the RAD50 gene, results from an A to G substitution at nucleotide position 1483. The serine at codon 495 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
