Uncertain significance — the classification assigned by Ambry Genetics to NM_021930.6(RINT1):c.1038C>G (p.Asn346Lys), citing Ambry Variant Classification Scheme 2023: The p.N346K variant (also known as c.1038C>G), located in coding exon 8 of the RINT1 gene, results from a C to G substitution at nucleotide position 1038. The asparagine at codon 346 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
