Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.1445G>A (p.Ser482Asn), citing Ambry Variant Classification Scheme 2023: The p.S482N variant (also known as c.1445G>A), located in coding exon 11 of the RECQL gene, results from a G to A substitution at nucleotide position 1445. The serine at codon 482 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
