Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.1442_1443del (p.Ala480_Leu481insTer), citing Ambry Variant Classification Scheme 2023: The c.1442_1443delTA pathogenic mutation, located in coding exon 12 of the CHEK2 gene, results from a deletion of two nucleotides at nucleotide positions 1442 to 1443, causing a translational frameshift with a predicted alternate stop codon (p.L481*). This variant was reported in 1/60,466 breast cancer cases and in 0/53,461 controls (Dorling et al. N Engl J Med. 2021 02;384:428-439). ( et al. N Engl J Med, 2021 02;384:428-439). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
