NM_004336.5(BUB1):c.1424T>G (p.Phe475Cys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F475C variant (also known as c.1424T>G), located in coding exon 13 of the BUB1 gene, results from a T to G substitution at nucleotide position 1424. The phenylalanine at codon 475 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
