Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_004612.4(TGFBR1):c.1424G>C (p.Trp475Ser), citing Ambry Variant Classification Scheme 2023: The p.W475S variant (also known as c.1424G>C), located in coding exon 9 of the TGFBR1 gene, results from a G to C substitution at nucleotide position 1424. The tryptophan at codon 475 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
