NM_001035.3(RYR2):c.14219C>G (p.Ala4740Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A4740G variant (also known as c.14219C>G), located in coding exon 99 of the RYR2 gene, results from a C to G substitution at nucleotide position 14219. The alanine at codon 4740 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
