Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004281.4(BAG3):c.1402G>A (p.Val468Met), citing Ambry Variant Classification Scheme 2023: The p.V468M variant (also known as c.1402G>A), located in coding exon 4 of the BAG3 gene, results from a G to A substitution at nucleotide position 1402. The valine at codon 468 is replaced by methionine, an amino acid with highly similar properties. This variant was reported in an individual with familial dilated cardiomyopathy (DCM), and it segregated with disease in the only affected family member who had genetic testing (Villard E et al. Eur. Heart J., 2011 May;32:1065-76). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
