Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.1400C>T (p.Ser467Phe), citing Ambry Variant Classification Scheme 2023: The p.S467F variant (also known as c.1400C>T), located in coding exon 13 of the CBS gene, results from a C to T substitution at nucleotide position 1400. The serine at codon 467 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
