Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.1527G>T (p.Leu509Phe), citing Ambry Variant Classification Scheme 2023: The p.L463F variant (also known as c.1389G>T), located in coding exon 14 of the KIF1B gene, results from a G to T substitution at nucleotide position 1389. The leucine at codon 463 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
