Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.1031G>T (p.Cys344Phe), citing Ambry Variant Classification Scheme 2023: The p.C344F variant (also known as c.1031G>T), located in coding exon 2 of the PALLD gene, results from a G to T substitution at nucleotide position 1031. The cysteine at codon 344 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
