NM_001378969.1(KCND3):c.1376C>A (p.Thr459Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T459N variant (also known as c.1376C>A), located in coding exon 4 of the KCND3 gene, results from a C to A substitution at nucleotide position 1376. The threonine at codon 459 is replaced by asparagine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
