NM_001166108.2(PALLD):c.1375G>T (p.Val459Phe) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V459F variant (also known as c.1375G>T), located in coding exon 6 of the PALLD gene, results from a G to T substitution at nucleotide position 1375. The valine at codon 459 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
