NM_002691.4(POLD1):c.1363G>T (p.Val455Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the POLD1 gene (transcript NM_002691.4) at coding-DNA position 1363, where G is replaced by T; at the protein level this means replaces valine at residue 455 with leucine — a missense variant. Submitter rationale: The p.V455L variant (also known as c.1363G>T), located in coding exon 10 of the POLD1 gene, results from a G to T substitution at nucleotide position 1363. The valine at codon 455 is replaced by leucine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.004% (greater than 25000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of p.V455L remains unclear.
