Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.1360A>T (p.Arg454Trp), citing Ambry Variant Classification Scheme 2023: The p.R454W variant (also known as c.1360A>T), located in coding exon 12 of the TSC2 gene, results from an A to T substitution at nucleotide position 1360. The arginine at codon 454 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
