NM_001430.5(EPAS1):c.1357A>G (p.Ser453Gly) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S453G variant (also known as c.1357A>G), located in coding exon 10 of the EPAS1 gene, results from an A to G substitution at nucleotide position 1357. The serine at codon 453 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
