NM_016203.4(PRKAG2):c.1349A>G (p.Asn450Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N450S variant (also known as c.1349A>G), located in coding exon 12 of the PRKAG2 gene, results from an A to G substitution at nucleotide position 1349. The asparagine at codon 450 is replaced by serine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, p.S450 is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
