NM_000179.3(MSH6):c.1342G>A (p.Gly448Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G448R variant (also known as c.1342G>A), located in coding exon 4 of the MSH6 gene, results from a G to A substitution at nucleotide position 1342. The glycine at codon 448 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
