Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.1331T>A (p.Val444Glu), citing Ambry Variant Classification Scheme 2023: The p.V444E variant (also known as c.1331T>A), located in coding exon 10 of the RECQL gene, results from a T to A substitution at nucleotide position 1331. The valine at codon 444 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
