Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003242.6(TGFBR2):c.1331A>G (p.Gln444Arg), citing Ambry Variant Classification Scheme 2023: The p.Q444R variant (also known as c.1331A>G), located in coding exon 5 of the TGFBR2 gene, results from an A to G substitution at nucleotide position 1331. The glutamine at codon 444 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
