Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005359.6(SMAD4):c.1330C>G (p.His444Asp), citing Ambry Variant Classification Scheme 2023: The p.H444D variant (also known as c.1330C>G), located in coding exon 10 of the SMAD4 gene, results from a C to G substitution at nucleotide position 1330. The histidine at codon 444 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
