Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.1325T>A (p.Ile442Lys), citing Ambry Variant Classification Scheme 2023: The p.I442K variant (also known as c.1325T>A), located in coding exon 9 of the RAD50 gene, results from a T to A substitution at nucleotide position 1325. The isoleucine at codon 442 is replaced by lysine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
