NM_001386125.1(OBSCN):c.16061A>C (p.His5354Pro) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H4397P variant (also known as c.13190A>C), located in coding exon 50 of the OBSCN gene, results from an A to C substitution at nucleotide position 13190. The histidine at codon 4397 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
