Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001082486.2(ACD):c.1057C>G (p.His353Asp), citing Ambry Variant Classification Scheme 2023: The p.H439D variant (also known as c.1315C>G), located in coding exon 10 of the ACD gene, results from a C to G substitution at nucleotide position 1315. The histidine at codon 439 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
